We urgently need to upgrade our existing CCD camera (Ultrascan4000) from Gatan and the optimal upgrade will be the K2 Summit camera as outlined below. The current camera can not perform in terms of obtaining high resolution structures based on cryoEM (only resolution above 10Å can be obtained). At Aarhus University we have a high end Titan Krios 300 kV electron microscope equipped with a Cs corrector and with the upgrade this set-up will be in the top 10 in the world and allow researches at Aarhus University and Denmark to perform cutting edge research and obtain high resolution (better than 4Å) structures of macromolecules based on cryoEM data. The Gatan K2 Summit camera has been benchmarked against competing cameras from other companies numerous times during the last 5 years. Benchmarking was done by independent research groups and there is a clear consensus that the K2 Summit camera performs best regarding the important parameters needed for obtaining high resolution structures. The unmatched frame rate of this direct detector allows electron counting modes to improve both contrast and resolution for large and small molecules alike. Recently, high resolution structures of a number of small membrane proteins and protein complexes have been determined using the K2 detector and these include the gamma secretase (1), the TRPV1 (2) and TRPA1 (3) channels, and the voltage-gated calcium channel CaV (4). The current highest resolution structure by cryoEM at 2.2Å of beta galactosidase (5) was also determined using the K2 detector. As there is an internationally highly recognized membrane protein structure community at Aarhus University, there is an urgent need for upgrading the existing camera to a K2 detector in order to secure a competitive edge for this community. The upgrade will of course benefit the entire structural biology community at Aarhus University and in Denmark.
