Section II: Object
Acquisition of a LC-MS
The instrument is going to be used to ongoing research projects and will be part of a major makeover of the LC-MS facilities.
The instrument is going to be used to ongoing research projects and will be part of a major makeover of the LC-MS facilities. The Danish food industry will also be using the instrument in collaboration with KU.FOOD under the FoodHay (National Research Infrastructure) umbrella.
Section IV: Procedure
- The works, supplies or services can be provided only by a particular economic operator for the following reason:
- protection of exclusive rights, including intellectual property rights
For a mid-range high throughput LC-MS system it is often the combination of soft- and hardware that creates the advantage. The IMPACT II from Bruker has the following three unique advantages that makes it an ideal instrument for our FOODOMICS research:
High throughput. The system can perform rapid and highly sensitive measurements via a patented ion optics technology based on in-flight adjustment on the nanosecond time scale which is important for achieving very high sensitivity at ultra-high resolution. The sensitivity is larger than 1 000:1 for 1 picogram (1.0E-12 gram) of the standard compound ‘reserpine’ on the column and the acquisition speed can be up to 60 Hz in MS and MS/MS without affecting the resolution or mass accuracy. The technology facilitates a great dynamic range without any compromise on the measurement speed which makes it unique for foodomics applications where investigated biological sample matrices are very different. The system further have the scientific reputation of being extraordinary robust for reproducible measurements of large sample numbers (co-horts) and is being acquired by the new globally leading phenome-centers.
Identification. The instrument provides a very accurate measurement of the isotopic intensities (0.8 ppm in MS and MS/MS using internal reference mass correction). This is an important orthogonal information to the accurate mass information generated by the instrument for confidently identifying compounds. Fully automated isotope matching for both MS and MS/MS. This unique feature allows identification of food derived unknown compounds.
Software. The second generation LC-MS software allows for data-sharing (import and export) with all established formats, enabling data evaluation in 3rd parts tools, e.g. for advanced statistics, pathway mapping or network analysis. It also allows direct annotation of metabolites, database search, tools for in-silico structure elucidation, build-up of libraries for UV spectra, etc. Bruker’s unique LC-MS software also integrates important databases such as The Bruker MetaboBASE Personal Library, The Bruker HMDB (Human Metabolome Data Base) Metabolite Library and The Bruker-Sumner Plant MetaboBASE library. The MetaboScape software includes discovery and exploration of new compounds, metabolic pathways and statistical comparison including support for hypothesis and experimental design for non-targeted experiments.
