Acquisition of a BD LSR Fortessa 20X (2B/3R/6V/5YG/2UV)
At BRIC we have four analysers (BD Calibur, BD LSR II, BD Celesta and BD Accuri) and among these the BD LSRII is the only instrument which can detect 20 parameters, rest are between 4-10 parameters.

The LSR II is about to retire soon and needs a replacement as soon as possible. Our committee chose BD LSR Fortessa 20x, which has similar configuration as LSRII with better electronics and users don’t need to change their existing panels, which constitutes a major advantage. The machine will be placed at the Core facility and made available to use by BRIC, KU and other academic and non-academic users.

- absence of competition for technical reasons
BD Biosciences is a major player with flow cytometry and all our instruments is supplied by this vendor. The machine operation and the software are similar between LSRII (our currently high-end analytical instrument) and the Fortessa 20x, thus the 50 + users don’t need any additional training which will save us a lot of time and resources. No other manufacturers in the market provide the five-laser configuration to match our demand. More importantly, the configuration of Fortessa 20X is compatible with our sorters, so the users can plan cell sorting without changing their panel design and even the experiment can be moved between the cell sorter and the analyzer which is quite handy at times.
Technical specifics of key importance include:
1) fast plate sorting: less than 15 minutes per microtiter plate in high throughput mode using a 2-second acquisition, less than 44 minutes in standard mode using a 10-second acquisition;
2) high data acquisition rate: 40000 events/s with beads;
3) extremely high sensitivity due its unique fluorescence detection technology with reflective transfer legs for color decimation
A key feature of the BD LSR Fortessa 20x is its sensitivity which is in part due to its fluorescence detection technology covered by US patents 6683314 and 7129505.
