Explanation:

There is a requirement of a motorized fluorescent microscope that allows automatic image capture, including Z-stack acquisition, with high resolution, producing tridimensional images in a short time due to the nature of the samples to be analyzed. Due to the thickness of the samples and the number of optical sections to be captured, the high resolution camera Photometrics BSI Express (USB 3.2) will alleviate the problems of fluorescence decay.

For the workflow of our projects to be successful, it is essential that the microscope and the software are compatible with the NIS AR software platforms installed in several other computers in our laboratory, which are successfully used by our lab members and other laboratory users, as specified in our funded projects.

Moreover, the new NIS AR software offers an additional unique advantage since it now includes the new ‘Denoise.ai (artificial intelligence based noise removal)’ required for our applications. Changing the system, deconvolution software and analyses tools would come with a high risk of incompatibility of processing images and comparing them to prior experimental setups in a consistent and reproducible manner.

Also, the required NiE Nikon microscope perfectly fits in a prior laboratory space, since it will replace an existing Nikon microscope (which will be removed) with similar size. Other microscopes from different brands do not have the same size characteristics and pose the risk to require construction to build another laboratory with the proper ceiling separation and electricity requirements.

There are multiple manufacturers capable of manufacturing motorized fluorescence microscopes with Z-stack acquisition. Nikon is the only manufacturer providing the NIS AR software required for the successful continuation of our projects, as specified in our funded project, which now includes the new ‘Denoise.ai (artificial intelligence based noise removal)’ and Ramcon in the solely distributor of Nikon and its software in Denmark.
