We intend to procure the Nikon Yokogawa W1 spinning disc microscope. The product is intended for the Center for stem cell biology imaging platform, a microscopy facility with approx. 150 users in the next 5 years. The platform doesn't have a spinning disk microscope yet. Since the confocal microscopes are heavily overbooked, often for live cell imaging (14h per day, including weekends) and the demand is rising steeply a spinning disc microscope (ideal for live cell imaging) will help overcome this bottleneck.

IV.1) Description
IV.1.1) Type of procedure

  • The works, supplies or services can be provided only by a particular economic operator for the following reason:
    • absence of competition for technical reasons
Explanation:

— The Nikon Spinning disk microscope is equipped with a PFS (Perfect focus, which allows stable imaging of live cells, without z-drift for long time lapse series of up to 4 days.

— The Software is uniquely flexible and thereby allows us to image all our different specimen and numerous applications. The Job-structure in the Nikon Software, is allowing us to setup a huge number of different workflows for all our users in the facility and thereby enabling us to image with different features and parameters, all depending of the requirements of the according application/experiment.

— Nikon has an integrated water dispenser, which can be synchronized together with the job structure.

This together with JOBS, GA3 (analysis expansion) and water dispenser, we are able to make some of the most advanced experimental setups without having to program anything ourself. JOBS is the ‘programming’ interface in this case, which is easy to use with a drag and drop interface.

There is no other microscope with the Software/Hardware combination that allows us this level of flexibility in terms of using different samples/dishes/experiment parameters in a fully automated workflow.

Stage-up structure of the whole focusing unit.

This explains the stage up structure of the Nikon Ti2 microscope, that you are able to build a second level on the microscope and expand the Lightpath behind the microscope to include TIRF, SIM or STORM devices while having a confocal (point scanning or spinning disc) setup on the sideport. This means in praxis you are able to customize your microscope for the future and expand it to include super resolution in addition to the current setup. It is extremely important for us to purchase a system that is flexible and upgradeable. The requirements of the different groups/users are changing with time, which means that we need to purchase microscopes that can be adapted to the future needs. It is very likely that we will upgrade this system in the near future, so we are dependent of this opportunity.

This is a patent for the unique design of the Perfect Focus System (PFS) that Nikon is famous for.

The intended procurement is an expansion for live cell imaging of other existing microscopes for dedicated applications, well established at Danstem. This microscope is specially made for live cell imaging and will free some slots from the heavily overbooked confocals at Danstem.
