Aarhus University wants to buy equipment for the detection of macromolecular interactions under native conditions for research purposes. The aim of the research is to obtain highly accurate information on binding affinity and complex stoichiometry for macromolecular assemblies in a manner dependent on the hydrodynamic radii of the complexes.

II.2.4) Description of the procurement:

We want to conduct highly accurate determination of binding affinity, kinetics and complex formation between different types of biomolecules under native conditions. Most equipment available on the market relies on immobilisation of one of the interaction partners used when investigating binding affinity (on a surface or on beads). However, the process of immobilising a biomolecule comes with the risk of changing its conformation and function, and this is therefore not a suitable method to recapitulate biologically relevant interactions. We therefore want to procure a piece of equipment that solves this problem by using changes in hydrodynamic range to measure binding affinity, size and complex stoichiometry for biomolecules under native conditions. We also need the apparatus to be able to determine such values in a sample containing complex biofluids such as serum and plasma. The ability to investigate molecular interactions under native conditions is key to ongoing work at iNANO and in the Center for Cellular Pattern Signaling (CellPAT) and we can only do this with equipment that combines the following:

• absolute measurement of size;

• molecules/binding complexes/nanoparticles in the range from 0,5 to 1000 nanometers (diameter);

• in-solution measurement. No need for surface chemistry optimization and regeneration;

• quantification of proteins;

• quantification of nanoparticles;

• compatible with crude samples. Generates robust data at high-% content of plasma, serum, fermentation media, etc.
