The Proteomics platform at KU-Food is to be expanded by purchasing two LC-MS instruments. Proteomics at KU-Food are used to analyze proteins and modification of proteins in food. At KU-Food, there is an increased focus on proteins in food, especially in relation to creating a more sustainable food production. It is therefore necessary to purchase equipment that can be used to make a detailed characterization of food proteins, especially intact proteins. Furthermore, determination of the protein levels in microorganisms during fermentation of food using tandem mass tags is also a future request. There is also a need to be able to analyze a large number of samples for validation.

Explanation:

The Department of Food Science has for a number of years used high-resolution Orbitrap based mass spectrometry for qualitative and quantitative analysis of samples from different food matrices. It is therefore a request that the new systems are 100 % compatible with our existing system both in relation to established methods and data analysis. It is also a requirement that the new systems are compatible with each other in terms of methods, hardware and data analysis. There is also a desire that the HPLC systems to be used for the two mass spectrometry instruments are the same so that it is possible to use the same spare parts and that the competencies required for maintenance are the same.

At the Department of Food Science, University of Copenhagen, we have established a number of methods for characterizing peptides and proteins in various food matrices, among other things to investigate the proteolytic activity during food production, as it is non-specific cleavage, high-resolution MS is needed to ensure a significant identification of the peptides (> 200 000 at m/z 200). There is also a growing need to examine intact proteins from plants and milk with high mass accuracy in order to examine the protein composition as well as the degree of modification of the individual proteins. There is a great need to quantify modification at the amino acid, peptide and protein levels. In relation to food fermentation as well as determination of protein synthesis in bacteria from intestinal flora, there is a need to use 16 plex tandem mass tags (TMT) for relative quantification of proteins. This requires a very high resolution instrument in MSMS scan mode to be able to separate the fragment ions from the fragmentation of mass tags in the MSMS spectra and thus in the subsequent data processing make it possible to accurately calculate the relative content of all the proteins in the sample.

There is a need for a high-resolution instrument that requires orbitrap technology that is patented by Thermo
