The Maestro Pro MEA system allow non-invasive stimulation and continuous analyses of i.e. neurons, in high throughput formats and otherwise complex data are visualized real-time and readily analyzed for extraction. The system meets our need for an easy operable approach to functionally validate and characterize human stem-cell derived hypothalamic cultures and screening of large compound libraries on functional neurons. Service includes comprehensive repair on parts and repair labour.

Explanation:

To which purpose we will use the instrument:

We are studying neuronal stem cell derived human hypothalamic cultures and need a system to perform non-invasive investigations and functional characterization of the neurons. The cell model needs to be validated at several time points during the 50-day long differentiation protocol, to ensure optimal differentiation status and functional neurons. We need electrophysiology in a user-friendly high content screening format. It has to be possible to monitor the cells in 96-well format, as we, during the second phase of the project, will screen the cells for drug targets.

Which unique characteristics will be used:

We have identified the Maestro Pro system as a perfect match for our requirements. This system utilize cell culture plates with incorporated electrodes and offers a user-friendly electrophysiology approach for 6-, 24-, 48- and 96-well plates. Hardware-incorporated (769 simultaneously recorded low-noise) electrodes and environmental control enable stable short- or long-term recordings from or stimulation of selected or all electrodes. The instrument allow real-time data visualization and activity maps from individual/all electrodes. Complex neuronal data are easily analyzed and extracted as i.e. raster plots, compatible with microsoft excel and MATLAB, meeting our requirement of high content screening. An integrated humidity reservoir reduces evaporation and edge-effects.

Axion Maestro Z system is covered by the following patents: Axion European patent EP 3024920; EP 15806517.7. Axion US Patent US 9,290,75; US 9,279,801; US 9,329,168; US 8,639,329; US 9,684,008; and, Axion Japanese Patent 6,337,113.
