The procurement involves the establishement of an advanced digital holotomographic microscope that enables non-invasive 3D imaging of refractive index in living samples at high spatial resolution and under controlled temperature and oxygen conditions, in combination with 3D imaging of fluorescent markers in the samples (followed by 3D reconstruction).
The system will be used in basic research and will e.g. be used to map the optical properties of living cells (microalgae, cyanobacteria), and the spatial structure of cell organells (chloroplasts, mitochondria) in large unicellular protists, algae and bacteria.

TomoCube HT-2H is the only digital holotomografic microscope system on the market that combines 3D imaging of refractive index with both brightfield and fluorescence imaging of samples over a large, freely defined area that is not locked to a specific well-plate format. The system is also the only complete system on the market that has in-built combined imaging of 3D refractive index with 3D reconstruction of cell structures imaged as z-stacks of fluorescence following by 3D reconstruction.
The system also enables, as the only on the market, flexible use of filter combinations and external light sources for the fluorescence imaging in combination with the holotomographic imaging of refractive index.
The scientific use of the system requires the above-mentioned flexibility and ability to work with many different types of samples and with different configurations in terms of fluorescence channels, and type of sample containers.
