Acquisition of a Carl ZEISS Elyra 7
The Core Facility for Integrated Microscopy is an open access research facility that offers services to any scientists in need.
The Elyra7 from Carl Zeiss is equipped with two technologies in demand by CFIM users. High resolution live imaging of 3D volumes and dual color STORM imaging. With SIM square, the Elyra7 allows for very fast imaging with reduced photo-toxicity and 100nm x,y resolution. In addition, the Elyra7 is equipped with two cameras to perform simultaneous imaging of two colors in 3D for Single Molecule Localization Microscopy.

ELYRA 7 type eLS for SMLM & Lattice SIM is a high-end super-resolution fluorescence microscopy platform based on Lattice Structured Illumination Microscopy using a Lattice pattern (Lattice SIM) and Single Molecule Localization Microscopy (SMLM) combined in an economic way as both technologies share the same camera. The main two hardware elements are (i) an illumination module, which projects sequences of illumination patterns or activation beams onto the specimen and (ii) an appropriate detection camera (sCMOS for Lattice SIM and SMLM) which detects the generated fluorescence of the specimen. The registered images are subsequently computed to generate images of the specimen with a resolution that is superior to that given by the classical diffraction limit: up to 2-fold in all dimensions for Lattice SIM, and typically 10-fold laterally and axially for SMLM
ZEISS is the exclusive licensee of patent rights related to "Optical Lattice Microscopy", a technology invent¬ ed by Prof. Eric Betzig, now professor in the molecular and cell biology department and the Eugene D. Commins Presidential Chair in Experimental Physics at the University of California, Berkeley and senior fel¬ low at Howard Hughes Medical Institutes Janelia Research Campus.

With the ZEN Connect software the Elyra7 can be used for correlative microscopy with the other Zeiss CFIM microscopes. CFIM has many Carl Zeiss systems and the fact that they are operated with the same software interface and the images created with the different systems can be registered and correlated is of great advantage for CFIM users. In addition, CFIM has implemented a robust automatized deep-learning driven pixel and object classification pipeline (ZEN module Intellesis), which combined with the Image analysis module allows many CFIM users to have an efficient processing and analysis of their acquired data sets without format changes and the need to export their data to other software packages.
