The LEO Foundation Skin Immunology Research Center aims at understanding, preventing and treating skin diseases for the benefit of the individual patient. For this, the researchers at the center need access to a multiphoton microscope to perform intravital imaging on human skin transplants and several mice models. After a thorough screening and testing of the commercially available systems, the Leica STELLARIS 8 focus on DIVE/CRS was identified as the only possible microscope for the service. The intravital services will be offered open access to other research groups in collaboration with the Core Facility for Integrated Microscopy and the Department of Experimental Medicine at SUND.

Explanation:

Our research application requires 4D deep imaging of skin tissue with a sensitive, flexible and user-friendly system.

STELLARIS 8 focus on DIVE/CRS is a true confocal point scanning system with prism-based spectral detection design with computer controlled adjustable bandwidth for all fluorescence channels and offers enhanced detection efficiency. The extended detection range up to 850 nm allows the application and separation of an extended range of spectrally overlapping fluorophores. This will allow us to label up to five cell types/tissue structures simultaneously.

LIGHTNING allows for optimal extraction of image details and maximum resolution, thus expanding the imaging portfolio both in the classical range and beyond the diffraction limit. Features a tandem scanning system to provide highest axial resolution with galvanometric scanner, switchable between resonant and non-resonant mode. This will allow us to obtain high resolution 3D information on skin structural changes.

Equipped with Power HyD S detector, new standard for detection. All-round detector, high performance throughout the spectrum, two times increased photon detection efficiency in the blue-green range compared to standard multialkaline photomultiplier. Leica Si-based technology is compatible with TauSense in combination with the white light laser. This is very important to be able to detect cells expressing low levels of fluorescent proteins deep inside the tissue.

Multiphoton extension STELLARIS 8 DIVE provides imaging in 1mm depth or beyond. Variable beam expander and beamcatcher ensure stable performance and adaption to sample requirements. 4Tune is the only spectrally tunable non-descanned detector and provides spectral flexibilty through the visible spectrum with up to 4 spectral channels simultaneously and unlimited 1 | 10 sequentially. Equipped with Power HyD NDD, 4Tune enables TauSense with multiphoton excitation. Tausense with allow us to image several fluorescent proteins deep inside the tissue flexibly and fast.

The Leica STELLARIS 8 FALCON system provides fast FLIM (at confocal speed) with any imaging workflow. Quick and easy operation is ensured by the full integration within LAS X. STELLARIS 8 FALCON includes FLIM analysis tools, such as exponential fitting, FLIM phasor analysis, pattern fitting and FLIM-FRET analyzer. STELLARIS 8 FALCON allows to quantify fast cellular processes, like molecular interaction or molecular environment changes.
