Advanced sample preparation laboratory for soft matter microscopy
DTU Nanolab intends to procure a suite of instruments capable of performing a seamless workflow for sample preparation of biological and soft matter suitable for (3D) scanning electron microscopy analysis. The suite of instruments should be compatible with already existing equipment at DTU Nanolab and future proof for already planned extensions. The sample preparation flow should be capable of preparing biological and soft matter samples for analysis by means of electron microscopy while preserving the sample morphology without introducing artefacts.
Reliable and easy sample transfer between the instruments is of utmost importance to minimize the introduction of sample artefacts. The instruments will be used by students, researchers and external customers and should therefore be a proven and robust and safe design that enables training to a wide and varied user base. DTUs requirements can only be fulfilled by suite of instruments from Leica, that can only be purchased through Triolab.
The suite of instruments to be procured includes following instruments (all from Leica Microsystems):
- EM ICE: High pressure system for automatically freezing multiple aqueous samples while delivering optimal sample preservation with minimal artefacts.
- EM AFS2 + FSP: performs freeze substitution and progressive lowering of temperature (PLT) techniques and allows low temperature embedding and polymerization of resins in a closed system avoiding inhalation of toxic chemicals.
- EM ACE600: Fully automated, versatile high vacuum coater producing very thin, fine-grained, conductive metal and carbon coatings. Required for field-emission scanning electron microscopy and transmission electron microscopy applications.
The acquisition shall be fully compatible with already existing sample preparation equipment at DTU Nanolab including a Leica Microsystems EM UC7/FC7 ultramicrotome to ensure seamless workflow in the sample preparation procedures and thereby avoid potential artefacts from change of environment.
The EM ICE, EM AFS2+FSP, and EM ACE600 from Leica Microsystems fulfil the requirements for a suite of instruments compatible with existing instrumentations at DTU Nanolab, securing a seamless sample preparation workflow for Nanolab’s multiuser research infrastructure. DTU Nanolab requires the suite of instruments to fulfil the following criteria:
High Pressure Freeze System (EM ICE):
- Reproducible and reliable freezing. Fully automated freezing process including specimen transfer to LN2 guaranteeing optimal physiological state of the sample
- Automated sample storage for multiple freezing cycles and samples allowing for subsequent freezing cycles.
- Able to reliable freeze specimens up to 5mm in diameter
- Possibility for fully integrated optical or electrical stimuli during freezing process (field retrofit)
- Low LN2 consumption during operation from integrated LN2 dewar
Freeze substitution system (EM AFS2+FSP):
- Sample should be kept dry during transfer to minimize sample contamination
- Safe and efficient handling of reagents during operation
- Fast UV polymerization
- Reliable and reproducible freeze substitution process
Coating and Glow discharge system (EM ACE600):
- Reliable and reproducible coating processes with subsequential metal and carbon coating without breaking vacuum
- Multiple easy-to-change sources for various metal coating
- Uniform, controlled, thin film coating of large area samples (>50mm). High precision film thickness monitoring
- Protocol for large area glow discharge
- Possibility for coating of samples at cryogenic temperature and cryo transfer / environmental transfer to any SEM or analytical instrument (field retrofit)

With fully integrated automated processes and workflows, the Leica Microsystems suite offers the only viable solution to the requirement for future proof preparation of soft matter and biological samples for electron microcopy on the market. The systems have the robustness to afford training of, and usage to a user base with a broad range of competences, which is the user base DTU Nanolab as an infrastructure facility at DTU has.
DTU Nanolab’s focus on imaging and microanalysis of soft matter and biological samples will push the field of nanoscale imaging within microbiology and biotechnology with help from the unique robustness and reliability from the Leica Microsystem instruments necessary for successful development of the research field by a varied user base.
After extensive market research of all commercially available systems and suppliers, we have concluded that the suite of instruments from Leica consisting of the EM ICE, EM AF2+FSP, and EM ACE600 provides the only combination of system performance and support that can meet the technical requirements of DTU Nanolab and its user base as specified above.
The unique combination of reliability, reproducibility, ease-of-use, and mutual compatibility of the instruments as well as Leica Microsystems’ extensive applications experience in sample preparation equipment for soft matter and biological specimens compatible with electron microscopy set the standard in the market today that meet the technical specification required by DTU Nanolab. The suite of Leica instruments can only be purchased through Triolab A/S.
