The Department of Plant and Environmental Sciences at University of Copenhagen carries out research across all aspects of plant science, from the molecular level to the field. Several research groups work with plant metabolism and the chemistry of plant-microbe interactions, and the department hosts several platforms for metabolomics and imaging. The purchase of the timsTOF fleX-MALDI-2 is funded by the Novo Nordisk Foundation and will enable the establishment of a facility for mass spectrometry imaging for plant science.

The timsTOF fleX-MALDI-2 system with microGRID comes with both MS (mass spectrometry) and tims (trapped ion mobility spectrometry) detectors as well as a post-ionization laser (MALDI-2) for enhanced ionization and sensitivity, and in addition the capacity for imaging at a lateral resolution of 5 µm in square pixels to cover the entire sample. The instrument will be used for analyses of metabolites in plants and plant microbe interactions. Plants are much more metabolically complex than animals and it is paramount that instruments used for plant analyses have very high sensitivity and identification power, which will be provided by respectively MALDI-2 and the combination of accurate mass (qTOF-MS) and trapped ion mobility analyses.

Explanation:

In our work with imaging plant tissues, we require minimal pixel size (<10 µm) to reach single cell resolution, high identification power (qTOF-MS + tims) to identify and differentiate isobaric compounds in extremely complex mixtures and high sensitivity (MALDI-2) for detection of lowly abundant compounds and at high lateral resolution. The timsTOF fleX-MALDI-2 system with microGRID is currently the only system that combines all these features in a single instrument. We have tested the MALDI-2 function of the instrument (Murphy et al., 2022, Plant Physiology) to find ionization features typical of MALDI-2 in plant samples. Ion mobility has proven to enable separation of true isomoers and even chiral compounds, and the timsTOF-fleX MALDI-2 system is furthermore a combination instrument, with both ESI and MALDI ionization sources, that allows for ion mobility separation. Finally, the established analytical facility will have a wide range of users and a high throughput which will be supported by the Intellislides and SCiLS autopilot feature that allows automated analyses.
