Super-resolution microscopy for Single Molecule Localization Microscopy
The objective is to set up a new microscope allowing the nanoscopic observation and quantification of neuronal and synaptic proteins without acquisition bias.
Dr. Chao Sun’s team of DANDRITE, Aarhus University, wishes to acquire a new microscope to carry out multicolor super-resolution based on the new technology dynamic illumination. This microscope is a development instrument that will allow the imaging of living or fixed biological systems using various optical modalities giving access to spatial resolutions around 20 nm.
The objective is to set up a new microscope allowing the nanoscopic observation and quantification of neuronal and synaptic proteins without acquisition bias. With this new microscope, our laboratory, a pioneer in these super-resolution techniques, will be equipped with a customized instrument unique in the world in order to better understand the local synaptic protein function and its impact on synaptic function.
General characteristics
- Super-resolution microscopy able to perform all Single Molecule Localization Microscopy such as PALM, PAINT and dSTORM with acquisition and analysis software.
- Super-resolution microscopy able to perform simultaneous multi-color imaging without compromise in speed, FOV and resolution.
- Super-resolution microscope able to perform 3D with magnified astigmatism.
- Wide field and homogeneous illumination thanks to the patented ASTER technique for adaptable scanning for top-hat excitation scanning renderer.
- Adjustable TIRF illumination for optimum adjustment of the illumination angle.
This instrument is designed to realize nanoscopy applications based on the single molecule localization approach. The objective is to reach a localization accuracy of 15x15x25 nm in XYZ.
- The SMLM instrument is coupled with an Olympus Evident inverted microscope and use only one camera port (the other ports remaining available for the coupling of other imaging modalities).
- The module can provide different illumination methods: EPI, TIRF, HiLo.
- The illumination must be homogeneous in the different illumination modes: EPI, TIRF, HiLo to perform quantification on the entire useful field. Specify the size of the field for a 95% illumination homogeneity (intensity difference between the center and the edge of the field).
- The field of view of the imaging should be as large as possible. A field of at least 150x150 µm2 with a 100x objective will be preferred in order to visualize the expansive dendritic and axonal arborization of neurons in culctures.
The instrument must include the following lasers. The laser power especially for 561 and 640 nm is important to ensure a high irradiance on the sample in full field for dSTORM applications.
o 405 nm with a minimum output power of 50 mW.
o 488 nm with a minimum output power of 40 mW.
o 561 nm with a minimum output power of 300 mW.
o 640 nm with a minimum output power of 500 mW.

The instrument must allow the Sun lab to carry out microscopy projects at a resolution of up to 20 nm, and this in a robust manner in three dimensions and in several colors. This is necessary to visualize the relative three-dimensional arrangement and copy numbers of synaptic proteins.
The instrument must be a stand-alone module combined with advance inverted microscopy frame, and that integrates high power laser illumination, and allows the use of fast and sensitive cameras for image acquisition.
The functionalities (simultaneous multicolor 3D SMLM, the largest homogeneous Field of View in TIRF), the type of equipment (stand-alone module for Single Molecule Localization Microscopy), and the integrated SMLM analysis software packages limit the offer to the SAFe360 module proposed by the Abbelight company.
