Section II: Object
Delivery of a cell sorter
Steno Diabetes Center Aarhus (STCA) wishes to purchase a 5-laser BigFoot cell sorter. There is a need to be able to sort live GMO classified cells as well as infectious cells, therefore the sorter must be within a GMO class II biosafety cabinet. Low noise is crucial, since STCA have to work and teach in the room the sorter is to be placed in. In addition, STCA have a need to sort many plates (96, 384 and 1536 well plates) with a high speed to improve cell viability and complicated combinations of fluorochromes.
Aarhus Universitetshospital
All of the following requirements are needed:
1) The sorter must be inside a GMO class II biosafety hood
2) The sorter must have the following spatial separated lasers to cover our need of fluorescent probes (few examples for each laser line is given):
349-355 nm: All the BUV dyes, Hoechst, DAPI, Qdots
405 nm: All the BV dyes, Super bright violet dyes and super bright dyes
488 nm: FITC, GFP, SYTO9, AF488, PerCP, PerCP-Cy5.5, BB700
561 nm: PE, PE-tandems, RFP, AsRed, mCherry, tdTomato
637-642 nm: APC, AF647, Draq7, AF700, nIR live/dead, APC-Fire750
3) The sorter must be a jet-in-air sorter to accommodate the need for high-speed cell sorting
4) The cell sorter must be able to analyze the full spectrum of the fluorophores and use spectral unmixing for sorting and in addition, it must also be able use conventional compensation
5) High-speed multi-stream plate sorting for 96- and 384-well plates
6) The sorter must be able to sort into 1536-well plates for single cell sorting
7) The sorter must be able to sort 6 populations simultaneously into both 1.5 ml and 5 ml tubes to avoid loss of sample (especially needed when sorting patient samples)
8) The sorter must be able to perform straight down sorting for both 96-, 384- and 1536 well plates (deflecting the waste stream)
9) Collection tubes must be temperature controlled
10) Flexible choice of nozzles to accommodate our different users. We need nozzles covering these diameters: 70, 100, 120 and 150 µm
11) Due to working environment the instrument including the biosafety cabinet must be silent (less than 60 db).
12) It must be possible to upgrade the sorter with more lasers to accommodate future experiments.
Section IV: Procedure
- The works, supplies or services can be provided only by a particular economic operator for the following reason:
- absence of competition for technical reasons
SDCA have checked the marked among all suppliers making fluorescent activated cell sorters. The only instrument on the market there can fulfill all the technical requirements is the 5-laser BigFoot cell sorter from ThermoFischer which SDCA aim to buy.
