Acquisition of a Zeiss Axio Observer 7
Department of Neuroscience at the Faculty of Health and Medical Sciences.
A high-end epifluorescence microscope specialized for live cell imaging. More specifically, it will primarily be used for imaging fluorescent (and to an extent FRET) biosensors in live cells during pharmacological manipulation. It will be heavily used by the group, sometimes for whole days for a full week. Data from this microscope will be combined with data generated from other Zeiss microscopes.

- Additional deliveries by the original supplier ordered under the strict conditions stated in the directive
It has some specific requirements that in combination are unique to Zeiss: Excitation for 385, 430, 475, 555, 590, 630, 735 nm in combination with dual filter wheel with 8 positions for specific beam splitter and emission filters (up to 64 filter combinations). Simultaneous emission reading with one camera of two channels Green/Red (~510/~620 nm) and preferably up to five channels (~425/~510/~600/~680/~780 nm). Upgradable for Water Auto-Immersion usable with all water-objectives in the nosepiece. Acquisition enabled by motorized components of large area images or z-stacks. Z-stack and enhanced depth of focus functionality. Minimum increment of 10 nanometers. Automatic scanning of predefined sample areas. Supports focus correction maps, stitching and shading correction. Compatible with software autofocus. The autofocus most work with transmitted, reflected and fluorescence light with same camera interface. Definition of an autofocus region in the camera.
The intended procurement complements the existing Carl ZEISS systems in our facility. To an extent, operation and data analysis will be streamlined between the intended procurement and existing systems. Experiments and data from microscope systems must be fully compatible and comparable with the same data format and ability.
In particular: Image acquisition and processing, user interface and control of the microscope systems and components, extensive acquisition and analysis. Acquisition of images according to a position list. Systems hardware setup and same data and software format.
It is of great importance that the same accessories can be used between systems since we need them to be identical (not similar). Taken together, this will also avoid unnecessary costs associated with two different brands of systems. Finally, the core facility has an expertise in Zeiss microscopes.
