The Knudsen laboratory, is situated in the Department of biology, Section for Cell Biology and Physiology. The group works on pancreatic islets of langerhans, and use advanced imaging techniques as well as biochemical and physiological experiments to study islet biology and whole body metabolism.
The following specifications represent the requirements for the fulfilment of the scientific task. We need a spinning disk confocal microscopy (SDCM) imaging of live cells and tissue samples with the possibility of adding structured illumination microscopy (SIM) on the same platform, preferably using the same cameras and excitation source. The system needs to be flexible so the user can exchange, the disc to optimize/configure the CLSM for fast, high light throughput or deep confocal imaging and change all relevant filters (excitation(cleanup) filters, dichromatic mirrors and emission filters. The microscope must be versatile and be able to quickly scan large areas and have large field of view with a uniform illumination. This together with triggered acquisition for fast acquisition with minimal shift, is vital to insure fast data acquisition. This will be used to increase throughput of dynamic live cell imaging. Simultaneous imaging of two colors using a dual camara approach is also necessary for our goals and to avoid auto fluorescence, we also require a far-red laser @ around 750 nm.

The system has a large field of view (25mm diagonal - 17.7 x 17.7 mm) with a uniform 90% homogenous illumination over the entire 25mm FOV.
The system is designed with exchangeable disks to optimize/configure the CLSM for fast, high light throughput or deep confocal imaging.
The system is designed for the DeepSIM add-on, and has a seamless shift from confocal to super-resolution.
The system comes with a combined spinning disk confocal attached to one port on the microscope and uses the same cameras and illumination source for widefield and SDCM
The system comes with VIS – NIR lasers for multichannel imaging (408/473/545/635/750)
User exchangeable optical filters.
The system comes with software fully integrated and suited for automated image acquisition and analysis. The software can facilitate high speed automated flexible acquisition. As well as 2D and 3D tracking and powerful analysis and visualization tools.
Specifically designed for fast imaging with a high disc rotation speed (15K rounds pr minute).
This microscope will improve the imaging capabilities of the laboratory. The large field of view will allow for simultaneous imaging of several specimens at the same time, more than with the conventional field of view which with considerably improve speed and quality of data collection.
We work with 3d micro-organs and the versatility of the microscope with the ability to change filters and The disc to suit the acquisition mode deep imaging of structures or fast imaging of cellular processes is ideal for the work we do.
