The Department of Molecular Biology and Genetics wanted to buy a Exactive Plus Orbitrap Complete LC-MS/MS System. The research is focused on protein identification and characterization and includes a number of diverse project such as basic research of human biology, archaeology, optimization of industrial production, disease characterization and diagnostics. The laboratory are either responsible or take part in several courses on mass spectrometry. Currently the laboratory has Quadropole, Linear Trap Quadropole and Time of flight technologies. To support the educational part it is important to establish the most widely used analyzer technology (orbitrap).

The system is acquired to fulfil the following research and educational proposes

Establishment of Orbitrap technology in the laboratory for research and especially hands-on education of the technology

Complete bench top set-up with nanoLC system and MS instrument operated by the same software to handle LC system and data acquisition.

For protein identification and PTM characterization in complex samples. Data dependent acquisition must have a minimum resolution of 70 000 and <3 ppm accuracy.

Precursor trapping in minimum 4 sec to increase sensitivity and with sequential MSMS acquisition at a resolution of minimum 35000. Trapping and high resolution MSMS will be used for protein identification in very rare low amount samples. Among others these includes archaeological samples, palaeontological samples, laser capture from tissue (rare disease) and rare post-translational modifications.

Explanation:

 

The acquired system shall expand the capabilities of the current MS facility. The research is focused on protein identification and characterization and includes a number of diverse project such as basic research of human biology, archaeology, optimization of industrial production, disease characterization and diagnostics.

The laboratory are either responsible or take part in several courses on mass spectrometry. Currently the laboratory has Quadropole, Linear Trap Quadropole and Time of flight technologies. To support the educational part it is important to establish the most widely used analyzer technology (orbitrap).

The current set-up includes fast and quantitative set-ups, however to increase the output on low abundant and/or complex samples a high-resolution instrument with trapping capabilities is needed.

The system is acquired to fulfil the following research and educational proposes
