We require an automated system for cyclic staining (>75 targets per sample) and localization of proteins and RNA molecules in tissue sections. The system has to be compatible with using filter-based slides for subsequent handing using a Leica LMD7 laser capture microscope. Specifications include: 1) Ability to perform cyclical staining and imaging with directly conjugated primary antibodies on the same area of tissue on the same sample slide, facilitating the iterative staining and acquisition of hundreds of markers. 2) Photobleaching or controlled, enzymatically release of fluorochromes to ensure efficient and complete signal erasure and an almost endless cycle of iterative staining 3) Ability to label and image RNA and protein targets on the same tissue section (essential). 4) Comprehensive pre-tested RNA and antibody panels for multiplexing: same section protein and RNA 5) Ability to accommodate a wide range of sample types, including tissue specimens and adherent or suspended cells. 6) An open platform that can use “in house” reagents and is not limited to proprietary reagents. 7) A system that can be upgraded if future technology becomes available and a commitment to increase the applications of the platform further 8) Comprehensive sample and reagent management system (software) and unbiased data analysis (k-means clustering, UMAP, and t-SNE calculation) In our opinion, the only system on the market that conforms to all of the above specifications in the MACSima system from Miltenyi Biotec. 1. High Multiplexing Capability: MACSimaTM stands out as the only platform to image 400+ protein biomarkers in a walkaway fashion. This high multiplexing capability is essential and will enable researchers to adapt to the ever-growing field and needs to image more markers in the ever-growing spatial biology field. 2. Advanced Data Analysis: MACSimaTM provides the only advanced data analysis tools through MACS iQ View, offering researchers the ability to perform in-depth analysis and generate publication-grade figures. This sets it apart from competitors who offer only basic or third-party-dependent analysis options. 3. Versatile Sample Compatibility: MACSimaTM is compatible with various sample types, including tissues fixed with FFPE, PFA or Acetone. In addition, tissue microarrays, adherent cells, and suspension cells can be used, providing flexibility in experimental design for the facility. The photobleaching unit also enables us to image very auto-fluorescent tissue such as brain or lung slices. 4. Rapid Time to Data: MACSimaTM offers an impressively short time to data, with the ability to process and analyze high-content imaging experiments efficiently. This is due to the ease of optimisation with simple fluorochrome-conjugated primary antibodies and an inclusive data analysis software. Researchers can thus obtain valuable insights faster, accelerating the pace of discovery. 5.
