DTU will like to buy a BD FACSDiscover A8 5 Laser 78 detectors from BD Biosciences. The instrument will include the following : • Forward Scatter: Label-free brightfield imaging channel that measures light scattered at small forward angles by the cell as it passes through the laser interrogation point. • SSC Imaging: Label-free imaging channel measuring light scattered at 90° perpendicular to the laser path, capturing internal cellular complexity. • Lightloss Imaging: Measures the total photons lost from the laser beam due to both scattering and absorption by the cell, allowing Label-free brightfield imaging morphological assessment. • Channel 1: Fluorescence imaging (530/30nm range) • Channel 2: Fluorescence imaging (600/60nm range) • Channel 3: Fluorescence imaging (788/225nm range) These imaging channels enable spatial analysis of protein localisation, cellular interactions, and morphological characteristics that remain invisible to traditional flow cytometry. The system generates quantitative image parameters including: • Area and Perimeter measurements for size discrimination • Aspect Ratio and Eccentricity for shape analysis • Diffusivity for texture characterisation • Correlation and Distance for multi-channel co-localisation • Radial Moment for signal distribution analysis. Applications enabled by this technology include: • Identification of subcellular protein localisation (nuclear vs cytoplasmic) • Detection of cell-cell interactions and immunological synapses • Discrimination of budding yeast cells and cell aggregates • Analysis of phagocytosis and particle uptake • Label-free morphological study for downstream sorting. This document provides justification for the acquisition of a BD FACSDiscover A8 Cell Analyser through a manufacturer trade-in programme. The proposed acquisition represents a transformative upgrade to our flow cytometry capabilities, introducing proprietary imaging technology whilst maintaining seamless integration with our existing BD FACSDiscover S8 Cell Sorter infrastructure. The exceptional trade-in value offered by BD Biosciences, combined with the unique technological capabilities unavailable from any competing manufacturer, establishes this as a procurement of singular strategic value. 1. TECHNOLOGY JUSTIFICATION 1.1 BD CellView Image Technology The BD FACSDiscover A8 incorporates BD CellView Image Technology, a revolutionary camera-free imaging system that represents a fundamental advancement in flow cytometry. This proprietary technology employs orthogonal frequency domain multiplexing (OFDM) — adapted from wireless telecommunications, to generate high-resolution cellular images without traditional camera systems. Technical Implementation: The CellView system utilises radiofrequency-multiplexed excitation of fluorophores, allowing the instrument to capture complete cellular images using the same avalanche photodiodes (APDs) employed for spectral detection. This approach overcomes the fundamental throughput limitations of camera-based imaging systems, which cannot operate at the acquisition speeds required for high-throughput flow cytometry (typically 35,000 events/second in high-speed mode; 12,500 events/second in imaging mode). Relevancy for our laboratory The FACSDiscover A8 is the only spectral analyzer capable of meeting DTU's requirements for advanced cell analysis. Its integrated CellView imaging technology, unique to the BD FACSDiscover platform, enables real-time morphological assessment and spatial localisation of fluorescent signals within individual cells, capabilities unavailable on any competing spectral analyzer. This allows researchers to visualise protein co-localisation, identify cell-cell interactions such as immune synapses, and track nanoparticle uptake at subcellular resolution, providing biological context beyond intensity-based measurements alone. Critically, the A8 shares identical optical configuration, calibration standards, and software platform (FACSChorus) with our existing FACSDiscover S8 cell sorter. This standardisation means panels developed and validated on the A8 transfer directly to the S8 without requiring re-optimisation, a workflow impossible with any other analyzer-sorter combination. Researchers can perform panel development, compensation setup, and sample screening on the A8, then proceed to sorting on the S8 with full confidence in data reproducibility. BD's "perform as one" calibration philosophy ensures that experiments are transferable not only within our facility but across any FACSDiscover system worldwide, supporting multi-site collaborations and reproducibility standards. WORKFLOW INTEGRATION AND OPERATIONAL SYNERGY Identical Optical Configuration with BD FACSDiscover S8 As mention above the BD FACSDiscover A8 shares an identical optical architecture with our existing BD FACSDiscover S8 Cell Sorter, and this identical configuration is essential for our analyses process. The BD FACSDiscover A8 includes an integrated 96-well plate autoloader supporting both standard and deep-well plate formats. This automation enables walkaway operation for large-scale screening applications, substantially reducing operator time requirements whilst improving cost-per-data-point economics. 3COMPETITIVE LANDSCAPE AND SOLE-SOURCE JUSTIFICATION BD CellView Image Technology represents a proprietary innovation protected by multiple patents. No competing manufacturer offers comparable camera-free, high-throughput imaging capabilities integrated with full-spectrum flow cytometry. Alternative approaches to imaging flow cytometry (such as camera-based systems from other manufacturers) operate at acquisition rates 10-100 times slower than the BD CellView platform, rendering them unsuitable for high-throughput applications. None other platforms offer the CellView imaging technology that distinguishes the BD FACSDiscover A8. The superior imaging capabilities of the A8, and the identical optical architecture of BD FACSDiscover A8, with our existing BD FACSDiscover S8 Cell Sorter, which is essential for our analyses process, combined with the advantageous trade-in pricing, position this acquisition as exceptional value compared to any non-imaging spectral cytometer alternative. BD Biosciences has offered to accept our BD FACSymphony A5 SE as a trade-in towards the BD FACSDiscover A8, providing substantial value reduction. The trade-in credit substantially reduces the net acquisition cost to 723,000 DKK, representing a reduction compared to typical market pricing for comparable instrumentation.
