Description The heliXcyto has a number of specifications that meets the criteria for allowing single cell analysis for our research investigations, that also differentiates itself from an alternative technology that measures binding on a lawn of seeded cell population, the LigandTracer, specifically: 1) the ability to measure real-time binding on single cells in a fully automated device. 2) equipped with an advanced microfluidic system. 3) can detect two fluorescent signals (red and green) at a time. This offers the ability to use FRET assays as well. 4) offers persistent fluorescence recording during measurement (not intermittent) with up to 100 ms real-time data resolution. 5) measurement temperature can be regulated from 15-40°C on a reuseable chip, which is regenerated automatically. 6) equipped with an autosampler with 96-well plate capacity for automated throughput of ~ 10 runs (interactions) with only 30 min of hands-on time.

Other justification The heliXcyto machine is needed by researchers at Aarhus University to investigate advanced molecular analysis interaction on single cells in physiological conditions that requires this unique instrument. The heliXcyto provides real-time measurements of molecular binding events on single cells due to the unique cage capture feature on reuseable chips. Receptor-specific precision medicines rely on truly understanding interaction with cell receptor targets on single cells and the real-time binding events in their natural state. This is of particular importance for multivalent drug designs for tuning the affinity and avidity of interaction being developed at Aarhus University. Current gold-standard methods for measuring receptor-ligand binding events, however, such as surface plasmon resonance, only measure binding to the purified protein receptors on artificial surfaces. This fails to capture the complexity of intact cells, where receptor density, mobility and co-receptors shape the outcome of binding events, leaving a critical gap in therapeutic development and understanding of biological relevance. In addition, these technologies fail to perform biophysical analysis of membrane target molecules which cannot be isolated. Furthermore, the heliXcyto has a number of specifications that meets the criteria for allowing single cell analysis for our research investigations, that also differentiates itself from an alternative technology that measures binding on a lawn of seeded cell population, the LigandTracer, specifically: 1) the ability to measure real-time binding on single cells in a fully automated device. 2) equipped with an advanced microfluidic system. 3) can detect two fluorescent signals (red and green) at a time.
