Aarhus University will be investing in the OctedRED96e system. A necessity to support our research is to have a platform for measuring protein-receptor interactions in a fast and user-friendly manner. The proteins to investigate are typically modified versions of a “parent” protein for example conjugates, fusions, or mutated versions. To test the functionality of these protein-derivatives it is necessary to test any differences in receptor engagement evaluated by binding affinities (KD-values) obtained by biolayer interferometry (BLI) as previously performed by our BLItz® system for comparison. The Octet®RED96e system fit to this purpose.

Requirements for the system:

— compatible to the BLItz® system including biosensors and buffers,

— label-free and fluidics-free as well as based on bio-layer interferometry method in order to compare to previous results obtained with our BLItz® system,

— compatible with crude samples and non-destructive to samples,

— parallel measurements of multiple samples,

— user-friendly software for students to use.

The Octet®RED96e system will fulfill these requirements. The biosensors are compatible with both our BLItz® and the Octet®RED96e, and can therefore be regenerated and reused.

The Octet®RED96e will allow fast and unattended experiments and eliminate user-introduced errors during pipetting and shifting buffer tubes. The Octet®RED96e is automated and test and buffer materials are put in 96-well plates and put into the machine for pre-programmed measurements. This reduce errors and make the system more user-friendly to inter-user use, so students do not need to specialize. This will also limit experiment-to-experiment variations, as user-introduced errors are limited. Furthermore, it will allow faster readings as multiple biosensors can be used in parallel.

Explanation:

For biolayer interferometry (BLI) studies the Octet®RED96e System is, to our knowledge, the equipment that fulfills the requirement of being both micro fluidics-free and compatible to our previous BLItz® system, so the same buffers and biosensors can be used for our existing machine, the BLItz® system, and the Octet®RED96e System.
