Aarhus University aim to purchase manual UV fluorescence imaging system designed to identify protein crystal samples and clearly distinguish them from salt and small molecule crystals due to the intrinsic fluorescence arising from the chemical properties of protein (specifically tryptophan fluorescence). The ability to automatically screen large numbers of samples and clearly identify and discard false positives is paramount for our downstream experiments.

Aarhus University aim to purchase manual UV fluorescence imaging system designed to identify protein crystal samples and clearly distinguish them from salt and small molecule crystals due to the intrinsic fluorescence arising from the chemical properties of protein (specifically tryptophan fluorescence). The ability to automatically screen large numbers of samples and clearly identify and discard false positives is paramount for our downstream experiments.

We have analyzed the potential market as follows: First we have worked through the purchase-lot numbers at Aarhus University and found no providers that could supply what we need. We have contacted our network in the scientific field of macromolecular crystallography, and found one provider, who specialty in crystallography products, that can provide what we need.

They offer a JANSi UVEXm imagining system that has the capabilities we need. We have not found any other provider that can provide the needed combination of design with shared UV and brightfield optical paths into an imagining system with computational readout. This is a proprietary design feature of the UVEXm, and does not exist elsewhere.

We need an imaging system that fulfill the following specific requirements:

1. The system offered must be a high end imaging system capable of screening protein crystallisation drops (drop sizes 50 nL — 5 uL), in all industry standard tray formats.

2. The system must have a manual XY stage for the fine adjustment of crystallization trays.

3. Illumination must include a Transmitted light base unit with an LED light source.

4. The Transmitted light base must generate minimal heat and of long-life type.

5. The imaging system must be capable of imaging in bright field, polarizing and trp-fluorescence modes.

6. The imaging system offered must have minimum 5X and 15X objectives.

7. The imaging system offered must have cross-polarizer.

8. The imaging system must have UV excitation/emission optimized for Trp fluorescence, that form an integrated part of the system.

9. The imaging system must allow simple and rapid differentiation of protein crystals from salt crystals using trp fluorscence.

10. The system offered must be equipped with a computer system with the necessary attachments for integration with the imaging hardware, to allow the recording of images and storage of data.

11. The provided software must be able to superpose brightfield and fluorsecence images.
