II.2.4) Description of the procurement:

TWINSHIME is adapted from the SHIME reactor, a fully automated yet flexible and adaptable system representing the gastrointestinal tract of the adult human. SHIME consists of five successive reactors representing different parts of the human gut. TWINSHIME operates 2 such setups in parallel. It will be used only for scientific research to simulate gut microbiota from human fecal samples, and then to test the effect of food/dietary components, drugs or probiotics in that microbial ecosystem.

The contracting institution is the Novo Nordisk Foundation (NNF) Center for Basic Metabolic Research (CBMR), Faculty of Health Science, University of Copenhagen. The CBMR is a research center with a vision to be an international hub of excellence in interdisciplinary basic metabolic research. The mission of the CBMR is to discover fundamental processes that underpin the development of transformative diagnostics and prevention/treatment strategies for metabolic diseases.

Explanation:

SHIME® is a registered Trade Mark of ProDigest and University of Ghent (Belgium). In 2008, ProDigest obtained the exclusive worldwide commercial rights on the SHIME and has improved the design of the model over the last years.

Nowadays, SHIME in a computer-controlled system with unique capabilities:

— The only automated in vitro gut model available to purchase, with large enough volumes for 2 series of 5 different gut compartments (i.e. stomach, small intestine, ascending, transverse and descending colon),

— High flexibility for optimal experimental design: the 10 reactors can be used to simulate a study with 2 arms, 3 arms, only upper GI tract, 10 independent bioreactors (study for interindividual variability),

— The high flexibility comes also with an open software that allows any customized design,

— Possibility to perform long-term studies with longitudinal sample collection over several weeks,

— Unique M-SHIME configuration: by adding in-house developed mucosal microcosms it is possible to maintain luminal and mucosal microbiota, a unique feature of key importance in terms of gut microbial ecology.

Hence, in summary, it is a unique model in terms of technology and simulation of physiology/microbiology of the GI tract.
