Purchase of a Liquid Chromatography Mass Spectrometry (LC-MS) System
Aarhus University wants to buy a Liquid Chromatography Mass Spectrometry (LC-MS) system, to support our research.
Our Research Unit for Molecular Medicine (RUMM) intend to purchase a MS, which should deliver high mass resolution proteomics data and enable the necessary integration of data from previous and future studies.
Aim of the research:
Our Research Unit for Molecular Medicine (RUMM) has a long tradition of advanced analyses of proteins and their relation to diseases. During the last decade, RUMM has applied advanced mass spectrometry (MS). RUMM performs untargeted proteomics for the discovery of previously unknown disease markers, quantification of defect proteins (“mutant proteins”), mapping of protein modifications and targeted proteomics. MMF has based on its numerous untargeted proteomics studies performed on existing LC-MS system (nanoLC-Q-Exactive Orbitrap MS) created an in-house database. This database contains mass spectra and retention time data necessary for integration of data from different studies, performed on existing instrumentation and on the instrument intended to purchase, and for design of new targeted analyses using Parallel Reaction Monitoring, PRM. RUMM now intend to purchase a MS, which should deliver high mass resolution proteomics data and enable the necessary integration of data from previous and future studies.
Criteria for the analytical instrumentation:
— must be suitable for both untargeted or targeted type (PRM) of analyte screening and be capable of a broad range of qualitative and quantitative proteomics applications,
— should be capable of simultaneous quantitative and qualitative analysis, and there should be possibility to analyze data retrospectively for identification and quantification of new compounds,
— must have a minimum of 240 000 resolution at m/z 200 with less than 1 ppm mass accuracy. This will resolve combinations of isotopes and give confident structural confirmation,
— must generate data files of the same data format (RAW format) as the existing system of the customer, enabling analytical integration of the new MS system and the existing system (Q Exactive Orbitrap),
— must be capable of spectrum multiplexing and advanced signal processing alongside nanoHPLC compatible data acquisition speed for increased throughput and thus large scale complex proteomics samples,
— must be a high-resolution benchtop LC-MS/MS system with high-performance quadrupole precursor selection,
— the LC-MS/MS system should be from one producer and controlled by the same software package,
— must have HCD (Higher-energy C-trap dissociation) collision cell to allow high fragmentation efficiency and high quality MS/MS spectra making it suitable for the analysis of labile protein post translational modifications.

It is our assumption that there only is one company that manufactures such instrumentation and fulfill the above described criteria for the analytical instrumentation. it is a benchtop instrument, that has the required resolution, file format and HCD collision cell, and fulfill all our criteria.
