The acquisition concerns a NovaSeq 6000 for the Lundbeck Foundation GeoGenetics Centre, University of Copenhagen. The NovaSeq 6000 is the largest commercially available DNA short-read sequencing instrument.

II.2.4) Description of the procurement:

The purpose of the acquisition is to sequence ancient DNA from 5 000 samples, which poses capacity demands, and secondly, requirements for technical compatibility. As aDNA is degraded we require short-read sequencing. The only commercially available short-read DNA sequencing instrument that meets our demand (>7.4 x 12 read-pairs in 2,5 years) is Illumina’s NovaSeq6000 combined with S4 flowcells. No other vendor is offering a similar capacity in a single instrument.

As the future generated data must be perfectly compatible with the colossal amount of data already produced inhouse over 8-9 years, we have tested the NovaSeq6000_S4, together with Illumina R&D, UK, and shown that NovaSeq data is compatible with existing other HiSeq platforms. Finally, the track record of Illumina’s instrument, service and support has worked fully to our overall satisfaction over many years.

The contracting institution is the Lundbeck Foundation GeoGenetics Centre, which is a newly founded Centre. Based on the previous Centre for GeoGenetics, the Centre has been world-leading in ancient human genomics and peopling of the continents for the last decade. The centre is headed by professor Eske Willerslev.

Explanation:

The only commercially available short-read DNA sequencing instrument that meets our demand for data output (7,4 x 12 read-pairs in 2,5 years) is the NovaSeq6000 combined with S4 flowcells. Each run is producing ≥1e10 reads/S4 flowcell from each of 2FCs in 48 hours. The sequencing data production is for 5 000 prehistoric samples that must be completed in 2,5 years. No other vendor is offering a similar capacity considering both data output/run as well as run time in a single instrument.

As the future generated data must be perfectly compatible with the colossal amount of data already produced in house over 8-9 years, we have tested the NovaSeq6000_S4, together with Illumina R&D, UK, and shown that NovaSeq data is compatible with existing other HiSeq platforms.
