II.2.4) Description of the procurement:

The IncuCyte S3 Live Cell Analysis System is a flexible assay platform that sits inside a standard tissue culture incubator and automatically acquires and analyses HD phase and fluorescent images of living cells, around the clock, for days, weeks, or months. Traditional “live cell assays” begin with living cells, but the assay process destroys the cells. With the IncuCyte S3, cells are measured in situ in a physiologically relevant environment. Continuous analysis ensures you don’t miss a relevant response or interaction. 96 and 384 well assay formats enable rapid optimisation of experimental variables and treatment conditions, such as seeding densities, concentrations or coatings. Because the IncuCyte S3 sits inside the incubator there is no disturbance of cells caused by bringing the cells to the instrument and no complicated environmental control systems required. Also, the optics within the system move to the plate, allowing the cells to remain stationary, and so the system is perfect for sensitive and non-adherent cell types. The optical system includes 4x, 10x and 20x objectives mounted on a motorised turret. The IncuCyte S3 is compatible with a wide range of culture vessels (96 and 384 well plates and culture flasks) and applications including Apoptosis, Cytotoxicity, proliferation, Neurite Analysis, Tumour spheroid analysis, scratch wound migration and invasion, Chemotaxis, immune cell clustering, Immune Cell Killing, Phagocytosis, Angiogenesis, dilution cloning, transfection efficiency and reporter gene activity. Many of these assays are run with the standard software although some require extra software modules to be purchased. The In-cuCyte S3 allows for simple and flexible experimental set up with a guided interface allowing for easy set up of automated acquisition parameters. Multiple users can schedule experiments using different acquisition frequencies and magnifications and up to 6 separate experiments can be run at the same time. The PlateMap function allows for experimental conditions to be tracked from set up through to analysis. Finally, the IncuCyte S3 can be controlled from any networked location using unlimited, free licenses for the whole facility.

Explanation:

There are few other instruments providing a similar plate-reader-based setup, however, they do not meet some of the technical specifications.

Some allow imaging of live cells with a variety of objectives and fluorescent channels, however they offer no environmental control in the instrument and require the use of an external incuba-tor. This necessitates the movement of plates to the instrument leading to disturbance of the cultures, making it difficult to use for suspension cell cultures. Most systems can only hold one vessel at a time, significantly limiting throughput and requires large amounts of additional bench space.

IncuCyte S3 Live Cell Analysis System comes with several advantages compared to the alternatives included in the evaluation. Firstly, it provides a possibility to reproduce and compare the results from leading experts in different laboratories around the globe using the same system. Secondly, it enables strict environmental control, can hold numerous culture vessels at the same time, which enables a higher throughput, and is very suitable for suspension cell cultures allowing them to lay in the incubator space an limits the movement which could disturb data collection. The unit itself is placed in the incubator, minimising the footprint in the laboratory. Additionally, it comes with several software modules which can replace some of the other machines and provide multiple solutions in a single unit.
