Acquisition of a inVia Qontor Confocal Raman Microscope
This product will be used for the investigation and visualization and quantification of fine details in single cells from different lines and under action of distinct chemotherapy agents.
This product will be used for the investigation and visualization and quantification of fine details in single cells from different lines and under action of distinct chemotherapy agents. In this project, morphological changes in the cells will be reported together with information regarding the biomolecular specificity of the chemotherapy agents. For that, we will follow modifications in fingerprinted vibrational modes in response to drugs targeting. The high wavenumber spectra will provide information on changes in lipid composition, while highly specific molecular markers will be used to determine protein content. Cellular response to the drug treatment will be followed over time. For that an unique incubation chamber adapted to the microscope is needed.

LiveTrack™: An auto-focus system suitable for a spectroscopy system uses the same illumination beam as the exciting light for the spectroscopy. In contrast to other auto focus techniques available, Renishaw’s LiveTrack™ technology continually adjusts the height of the motorized microscope stage to maintain focus during viewing and scanning. This has many advantages especially to live cell research in the incubators where cell height, size etc. changes continuously.
StreamLine™ Plus Mapping: StreamLine Plus option provides high speed Raman mapping capability to the inVia system. StreamLine Plus uses optics within the inVia Raman microscope to illuminate a line on the sample. The inVia's motorised microscope stage moves the sample beneath the objective lens so that the line is rastered across the region of interest. Data are swept synchronously across the detector as the line moves across the sample, and are read out continuously. This enables very high speed Raman mapping as there is no dead time in the stage movement. StreamLineTM Plus maintains high sensitivity and high spatial resolution. StreamLine™ Plus is not limited to the field of view of the microscope objective as the sample is moved under the laser line. There are therefore no tiling effects often seen on other systems.
Confocal Optics: The optical design of the Renishaw inVia is inherently confocal. The system provides a confocal resolution of better than 2.0 μm when using x100 objective lens. The confocal arrangement uses a combination of the spectrograph slit and the active CCD area to obtain optimum performance. This is a very flexible solution that offers through software a continually variable confocal volume compared to the practical and limitations of using fixed pinholes. Systems using pinholes generally require regular realignment and refocusing to ensure optimum throughput and have a limited number of pinhole sizes available so the system is either highly confocal, with reduced throughput, or wide field. The Renishaw Easy Confocal™ system overcomes these problems and the degree of confocality can be easily adjusted via software to match the requirements of the sample under investigation. This is needed especially in the fast changing life science samples.
Synchroscan™: This is a unique ‘extended scanning’ facility for measurement of high resolution spectra with wider wavelength range than can be accommodated on a single CCD view, without any ‘stitching’ together of the spectra. It is ideal for measuring Raman spectra across the full range or Raman and photoluminescence spectra out to 1.1 micron. This is a really helpful technique for detecting unknown substances in lifescience as well archeological samples. The spectral resolution is continuously variable via software control of the CCD binning. The classical method is a step and repeat of the grating and, subsequently stitching the individual spectra together.
