Acquisition of a Zeiss LSM900 with Airyscan2

Prof Ole Kiehn’s group needs to perform fast super resolution imaging with a confocal microscope in order to image their samples. It will enable them to image large areas of hole brain sections with high resolution and fast. The confocal LSM900 will be equipped with the Airyscan detector, which allows us to image with lower light intensity and higher resolution, which is essential for many of the projects/applications to be performed.
Section IV: Procedure
- The works, supplies or services can be provided only by a particular economic operator for the following reason:
- absence of competition for technical reasons
With the airyscan the LSM900 allows for fast imaging with low photo-bleaching of large fields of view.
The LSM 900 with Airyscan 2 is the only laser scanning confocal microscope that allows us to acquire images with lower laser light intensity and higher resolution. It is a point scanning confocal with super resolution based on pixel reassignment detection technology. This will give an isotropic increase in resolution down 120 nm lateral and 350 nm axial. The new Multiplex mode provides greatly enhanced imaging speed (4x faster) than confocal imaging while providing better resolution and signal to noise. This allows gentle imaging at low laser power for high speed processes and also productive imaging of large fields of view in combination with Tile experiments with enhanced image quality.

Method for enhancing the resolution of a microscope during the detection of an illuminated specimen and a microscope for carrying out the method, wherein in a first position, an illumination pattern is generated on the specimen, the resolution of which is preferably within the range of the attainable optical resolution of the microscope or higher, wherein a relative movement, preferably perpendicular to the direction of illumination, from a first into at least one second position of the illumination pattern on the specimen is generated at least once between the detection and the illumination pattern with a step width smaller than the resolution limit of the microscope and detection and storage of the detection signals take place both in the first and in the second position.

High resolution scanning microscopy
Summary excerpt: a microscope for high-resolution scanning microscopy of a sample, with a spatial resolution which, taking account of the imaging scale, is at least twice as high as a full width at half maximum of the diffraction-limited
