II.2.4) Description of the procurement:

The acquisition provided is for 2 x 150 L customised pressure vessels for media supply and harvest of fermentation broth from currently onsite Frings Proreact15P. The vessels meet our requirements for SIP and CIP, full integration into existing system automation and physical piping and allowing for complex feeding of fermentation media through load-cells and flowmeters. Vessels include magnetic agitators, sensors for pH, temperature and pressure, ports for additions and including re-sterilisable sample ports. All contacting surfaces have hygienic design with surface roughness (Ra) < 0.8μm.

Customized tanks include installation and integration into Proreact15P, onsite water test and training for personnel.

In addition, future automation and needs will be supported by Frings for Proreact15P fermenter and 2 x 150 L customised pressure vessels as a shared service contract and preventive maintenance. Further common joined operational spare parts will be purchased from one sole vendor.

Explanation:

The Center for Biosustainability (CfB) at DTU provides interdisciplinary research with an emphasis on bioprocesses through microbial hosts. Our group, the Pre-Pilot Plant (PPP), already has capabilities for high throughput screening of various fermentation processes at smaller scales and is strengthening our capability for scale-up activities with our current 150 L fermenter — the Proreact15P. PPP is unique within CfB, as we collaborate with internal projects and external partners to leverage our unique pilot-scale equipment, to best simulate industrial scale conditions for fermentation and recovery operations. Which helps researchers assess and improve their microbial hosts through a design, build, test and learn cycle.

The addition of 2 x 150 L customised pressure vessels for our 150 L fermenter would expand our capabilities to: increase fermentation throughput, allow diverse feeding options or repeated fed-batch operations, realistic cleaning and sterilisation for components and all in effort to deliver reliable and scalable fermentations. In addition to scale-up activities, the customised pressure vessels will also increase throughput as media and harvested fermentation broth can be either prepared or stored in each vessel, while another is cleaned. To deliver our requirements and integrate seamlessly into our current Frings Proreact15P, we should continue with Frings for this current addition of 2 x 150 L customised pressure vessels. Their integration both physically and into the control systems is paramount for performance and safety. As the main Proreact15P control system will automatically issue commands to valves on all equipment for SIP, CIP, dosing of feed and other operations. To add, any future improvements of adjustments will be simpler and reliable with one supplier to rely on for upgrades. Learning shows, that interfaces handled by various vendors or parties are always prone for malfunction and problems.

In summary, Frings is the ideal vendor to continue the expansion of our Proreact15P system with the addition of 2 x 150 L customised pressure vessels due to the following:

— Frings built existing Proreact15P and are best positioned for seamless integration of physical piping and control system, along with automation of the customised pressure vessel;

— future adjustments or improvements of automation or physical equipment through one manufacturer/including operational spare parts;

— service of Proreact15P will expand to include customised pressure vessels, simplifying any technical issues or maintenance;

— Frings has demonstrable experience and knowledge with industrial projects;

— we are currently trained on Frings equipment and this will be an extension of Proreact15P.

It is therefore DTU’s assessment that only Frings can deliver the required goods and services.
