The Department of Biology at the University of Copenhagen is based on a legacy of outstanding researchers and is today one of the largest institutes at the University of Copenhagen. The institute has a very unique research and study environment with strong basic research education in biology, biochemistry, molecular biomedicine and bioinformatics.

II.2.4) Description of the procurement:

Intended procurement: Purchasing a motorized fluorescence microscope Nikon Ti2-E with environmental control chamber and Photometrics Kinetix camera.

Planned use: the microscope will be used in our ongoing projects which are strongly based on the quantitative analysis of cellular, organellar and extracellular pH, Ca2+, and other ion concentrations, in 2D as well as 3D cultures. These projects require rapid, live ratiometric imaging of low-level fluorescence, perfect focus, excellent noise subtraction, a large field of view, long working distance objectives, and complete environmental control.

Service context: Acquisition of the microscope, environmental chamber, camera and all accessories.

Explanation:

This setup will replace an existing Nikon setup in our lab, and the acquisition of a Nikon microscope means that we can reuse our expensive objectives in the new setup, saving substantial resources.

The proposed setup is specifically suited for our need for a very large field of view (FOV), and the Photonics Kinetix camera is unique on the market in having a 29 mm FOV.

The Nikon CFI S Plan Fluor LWD 20 XC, N.A. 0.70, W.D. 2.3 -1.3mm is unique in having an extremely long working distance, which is required for our studies of 3D cell cultures such as organoids and cysts.

The unique Clarify.ai artificial intelligence module in the NIS-Elements software allows feedback control of background noise – which will be essential for the precise, rapid measurements of low-level fluorescence.

RAMCON is the sole supplier with the right to sell Nikon microscopes in Denmark and also has the exclusive rights for the sale of the Photonics Kinetix large FOV camera. There are several exclusive patents on the setup: P2010-59631/ P2011-191671A and P2014-144121/ P2014-219690A.
