The department of biology intends to acquire a new 800 MHz NMR spectrometer with a cryogenic probe optimized for direct 13C and 15N detection. The equipment will be used for research in structure, dynamics and function of biomolecules, primarily proteins. In addition, the equipment will be part of an open NMR facility (cOpenNMR) to allow for easy access to high field NMR spectroscopy to scientists from all fields.

Explanation:

Direct 13C and 15N detection experiments can overcome limitations common to direct 1H detection in terms of resolution or relaxation properties. Yet, these experiments are often not feasible due to the low sensitivity of the X-channels of standard inverse probes. Bruker BioSpin is the only vendor of a cryogenically cooled 800 MHz probe optimized for 13C and 15N detection, with unique sensitivity (13C superior to 3000:1 on an ATSM standard sample and 15N superior to 150:1 on a formamide standard sample), making these experiments routinely accessible even at lower sample concentration. Furthermore, this probe will complement the 3 existing spectrometers (all optimized for 1H detection) and give an additional incentive for customers to use the cOpenNMR facility. Bruker cryoprobes also benefit from a unique 2H sensitivity, sufficiently high to reliably lock on 1 % D2O or less in order to shift exchange processes into more NMR beneficial regimes as well as the capability to measure and regulate the NMR sample temperature within the sample based on temperature dependent 2H signals. This will enable the acquisition of experiments at identical sample temperature independently of the spectrometer (600 MHz, 750 MHz, 800 MHz) and, more importantly, independently of varying RF load/RF heating in the sample (NMR thermometer).
