An additional inverted fluorescence confocal microscope is needed in order to manage the huge demand of users at CFIM for imaging their samples. It will enable us to image cells and tissues under natural conditions with an environmental control. It will be equipped with the Airyscan detector, which allows us to image with lower light intensity and higher resolution, which is essential for many of the projects/applications.

CFIM also has a need for a new stereo/zoom microscope which can acquire both overview images as well as more detailed images for both Brightfield and Fluorescence. The zoom microscope will be equipped with the ApoTome, which allows for optical sectioning.

Explanation:

With the ZEN Connect software the AxioZoom.V16 and the LSM 980 can be used for correlative microscopy to quickly acquire an overview image on the zoom microscope. The overview image can then be used for navigation on the confocal microscope. This will increase the productivity on the confocal microscope.

The LSM 980 with Airyscan 2 is the only laser scanning confocal microscope that allows us to acquire images with lower laser light intensity and higher resolution. It is a point scanning confocal with super resolution based on pixel reassignment detection technology. This will give an isotropic increase in resolution down 120 nm lateral and 350 nm axial. The new multiplex mode provides greatly enhanced imaging speed (8 x faster) than confocal imaging while providing better resolution and signal to noise. This allows gentle imaging at low laser power for high-speed processes and also productive imaging of large fields of view in combination with tile experiments with enhanced image quality.

The AxioZoom.V16 with the ApoTome.2 represents the most powerful combination of a large 16:1 zoom range with the highest basic NA 0.25 in the market. The ApoTome.2 is a device for structured illumination to acquire multichannel 3D fluorescence images of sections and whole animals. This provides images with increased resolution and increased spatial resolution. With this system, users at CFIM can acquire both macroscopic images of large samples as well as 3D images of e.g. cleared samples.
